Funktionen
Obwohl ADAMTS3 in die ADAMTS-Untergruppe der Prokollagenasen eingeordnet wird und in vitro Prokollagen proteolytisch schneidetcite-ref-2[2], ist ihr hauptsΓ€chliches Substrat der Wachstumsfaktor VEGF-C.cite-ref-3[3] Die Aktivierung von VEGF-C erfolgt, indem ADAMTS3 ein N-terminales Propeptid proteolytisch abspaltet. Dementsprechend liegt die Hauptfunktion von ADAMTS3 in der Regulation des LymphgefΓ€Γwachstums (der Lymphangiogenese). WΓ€hrend der Embryonalentwicklung ist ADAMTS3 fΓΌr die Entwicklung des LymphgefΓ€Γsystems essentiellcite-ref-4[4]. Ob ADAMTS3 auch im erwachsenen Organismus fΓΌr die Aufrechterhaltung und Neubildung von LymphgefΓ€Γen (z. B. bei der Wundheilung) notwendig ist, ist z. Z. noch ungeklΓ€rt, da andere Enzyme (z. B. Plasmin) VEGF-C zumindest in-vitro aktivieren kΓΆnnencite-ref-5[5].
Krankheitsrelevanz
Mutationen im ADAMTS3-Gen sind verantwortlich fΓΌr die Erbkrankheit Hennekam-Syndrom Typ IIIcite-ref-6[6]cite-ref-7[7].
